As for getting your design in, make sure you start a new theme... basically this means adding a new folder in 'wp-content/themes/' You can copy and paste everything from the 'default' folder into the new folder you have just created. From then on, all you have to do is edit each file, putting in the appropriate html, and also editing the stylesheet. If you know a bit of PHP, then you can use that to move things around on your page, or make information display differently. Once you're done with that, you just have to active your new theme in the control panel (wp-admin). I hope that gets you started, you can get lots of great help at the <a href="http://codex.wordpress.org/Blog_Design_and_Layout" rel="nofollow">Wordpress Codex</a> which got me on the right road to converting Cssdev over to 1.5.
